Application of TLC and GC-MS to the detection of capsaicin from hot peppers(Capsicum annuum)

Capsaicin was extracted from red hot pepper (Capsicum annuum) fruits and from commercial red hot pepper powder and separated on silica gel 60 plates with concentration zone (Merck); toluene-acetone-chloroform, 45 + 30 + 25 ( v/v ), was used as mobile phase. Detection was performed by exposure to iodine vapor. The spots assumed to arise from capsaicin were removed from the plate and the components were extracted with chloroform. The extracts were analyzed by gas chromatography combined with mass spectrometry.
The inner nuclear membrane protein Src1 associates with subtelomeric genes and alters their regulated gene expression

Inner nuclear membrane proteins containing a LEM (LAP2, emerin, and MAN1) domain participate in different processes, including chromatin organization, gene expression, and nuclear envelope biogenesis. In this study, we identify a robust genetic interaction between transcription export (TREX) factors and yeast Src1, an integral inner nuclear membrane protein that is homologous to vertebrate LEM2. DNA macroarray analysis revealed that the expression of the phosphate-regulated genes PHO11, PHO12, and PHO84 is up-regulated in src1Δ cells. Abstract An analytical method has been developed for the study of the elimination of 2,3,7,8-tetrachlodoribenzo-p-dioxin (TCDD) by lactation and its determination in various tissues of goat. C-28/C-30 oxidized cycloartanes from the leaves and twigs of Caloncoba dusenii Gilg

Abstract Four hitherto unreported cycloartane-type triterpenes (1-4) with C-28/C-30 oxidized methyl groups together with two known compounds (5-6) have been obtained from the leaves and twigs of Caloncoba dusenii Gilg. The compounds were isolated through extraction and silica gel column chromatography. Their structures and relative stereochemistry were determined by extensive use of spectroscopic techniques including IR, 1D and 2D-NMR as well as mass spectrometry. This is the first report of C-28/C-30 oxidized positions in cycloartanes from nature.

Abstract The thin-layer chromatography of 2-methyl-4-chlorophenoxyacetic acid, 4-chloro- o -cresol and 3-methyl-5-chlorocatechol and their pentafluorobenzyl derivatives has been studied on silica gel as adsorbent with 19 solvent systems. The best separation of the individual components occurred with toluene-benzene-acetic acid (2:2:1). Chloroform-diethyl ether-toluene (1:1:1) was suitable for the group separation of the pentafluorobenzyl derivatives.
Development and validation of RP-HPLC method for the quantitative estimation of αs1-genetic variants in goat milk

A high-performance liquid chromatographic (HPLC) method was developed and validated for separation and quantification of the most common genetic variants of as1-casein in goat’s milk, to evaluate the effect of as1-casein polymorphisms on casein content. Chromatography was carried out by binary gradient technique on a reversed-phase C8 Zorbax column and the detection was made at a wavelength of 214 nm. The procedure was developed using individual raw milk samples of Girgentana goats. For calibration experiments, pure genetic variants were extracted from individual milk samples of animals with known genotypes, considering that commercial standards for goat genetic variants were not available.…

Abstract The relationship between gas-liquid chromatographic (GLC) retention properties and R F values in thin-layer chromatography (TLC) with molecular connectivity indices, m X t , was investigated for a series of benzodiazepines using multiple correlation coefficients, standard errors of estimate, F -Snedecor function values and Student's t -test as the criteria for best equation selection. Abstract Separation of U(VI) and Th(IV) from some rare earths by TLC has been studied using silica gel H and silica gel H impregnated with ammonium nitrate as the stationary phase. The solvent mixture ethyl‐methyl‐ketone–tetrahydrofuran (6∶3, v/v) containing di-(2‐ethylhexyl)-dithiophosphoric acid was used as the mobile phase. When silica gel H impregnated with 2.5 M NH4NO3 was used as stationary phase, the resolution was much improved and the separation of rare earths from each other was also achieved. The tetrad‐effect was observed in variation of R*m versus the atomic number of rare earths.

A rapid and simple detection procedure of Aib-containing “peptaibol” polypeptide antibiotics (mycotoxins) in fungal culture broths is described which employs alkylsilica (Sep-Pak®) cartridges for a selective enrichment, and the utilization of the unusualα-aminoisobutyric acid (Aib) and amino alcohols as specific marker constituents, which are easily detectable in total hydrolysates. Preparative isolations from fermentation broths is facilitated by adsorber resin (XAD) column chromatography, and purification of crude isolates is achieved by silica gel- and sephadex LH-20 chromatography.
